Addendum: competing interests and the origins of SARS-CoV-2

In February, 2020, 27 public health experts co-authored a Correspondence in The Lancet (“Statement in support of the scientists, public health professionals, and medical professionals of China combatting COVID-19”),1 supporting health professionals and physicians in China during the early stages of the COVID-19 pandemic. In this letter, the authors declared no competing interests. Some readers have questioned the validity of this disclosure, particularly as it relates to one of the authors, Peter Daszak. In line with guidance from the International Committee of Medical Journal Editors, medical journals ask authors to report financial and non-financial relationships that may be relevant to interpreting the content of their manuscript.2 There may be differences in opinion as to what constitutes a competing interest. Transparent reporting allows readers to make judgments about these interests. Readers, in turn, have their own interests that could influence their evaluation of the work in question. With these facts in mind, The Lancet invited the 27 authors of the letter to re-evaluate their competing interests. Peter Daszak has expanded on his disclosure statements for three pieces relating to COVID-19 that he co-authored or contributed to in The Lancet—the February, 2020, Correspondence,1 as well as a Commission Statement3 and a Comment4 for the Lancet COVID-19 Commission. The updated disclosure statement from Peter Daszak is:

I have identified and recovered a deleted set of partial SARSCoV-2 sequences from the early Wuhan epidemic. Analysis of these sequences leads to several conclusions. First, the Huanan Seafood Market sequences that were the focus of the joint WHO China report (WHO 2021) are not representative of all SARSCoV-2 in Wuhan early in the epidemic. The deleted data as well as existing sequences from Wuhan-infected patients hospitalized in Guangdong show early Wuhan sequences often carried the T29095C mutation and were less likely to carry T8782C /C28144T than sequences in the joint WHO-China report (WHO 2021). Second, given current data, there are two plausible identities for the progenitor of all known SARS-CoV-2. One is proCoV2 described by Kumar et al. (2021), and the other is a sequence that carries three mutations (C8782T, T28144C, and C29095T) relative to Wuhan-Hu-1. Crucially, both putative progenitors are three mutations closer to SARS-CoV-2’s bat coronavirus relatives than sequences from the Huanan Seafood Market. Note also that the progenitor of all known SARS-CoV-2 sequences could still be downstream of the sequence that infected patient zero depending on the transmission dynamics of the first infections.

The fact that such an informative data set was deleted has implications beyond those gleaned directly from the recovered sequences. Samples from early outpatients in Wuhan are a gold mine for anyone seeking to understand spread of the virus.

Even my analysis of the partial sequences is revealing, and it clearly would have been more scientifically informative to fully sequence the samples rather than surreptitiously delete the partial sequences. There is no plausible scientific reason for the deletion: the sequences are perfectly concordant with the samples described in Wang et al. (2020a,b), there are no corrections to the paper, the paper states human subjects approval was obtained, and the sequencing shows no evidence of plasmid or sample-to-sample contamination. It therefore seems likely the sequences were deleted to obscure their existence. Particularly in light of the directive that labs destroy early samples (Pingui 2020) and multiple orders requiring approval of publications on COVID-19 (China CDC 2020; Kang et al. 2020a), this suggests a less than wholehearted effort to trace early spread of the epidemic.

Future studies should devote equal effort to going beyond the annotations in GISAID to carefully trace the location of patient infection and sample sequencing. The potential importance of such work is revealed by the observation that many of the sequences closest to SARS-CoV-2’s bat coronavirus relatives are from early patients who were infected in Wuhan, but then sequenced in and attributed to Guangdong.

More broadly, the approach taken here suggests it may be possible to learn more about the origin or early spread of SARSCoV-2 even without an international investigation. Minimally, it should be immediately possible for the NIH to determine the date and purported reason for deletion of the data set analyzed here, since the only way sequences can be deleted from the SRA is by an e-mail request to SRA staff (SRA 2021). In addition, I suggest it could be worthwhile to review e-mail records to identify other SRA deletions, which are already known to include SRR11119760 and SRR11119761 (USRTK 2020). Importantly, SRA deletions do not imply any malfeasance: there are legitimate reasons for removing sequencing runs, and the SRA houses >13-million runs making it infeasible for its staff to validate the rationale for all requests. However, the current study suggests that at least in one case, the trusting structures of science have been abused to obscure sequences relevant to the early spread of SARS-CoV-2 in Wuhan. A careful re-evaluation of other archived forms of scientific communication, reporting, and data could shed additional light on the early emergence of the virus.

The Debate over Origins of SARS-CoV-2

Caltech's David Baltimore, president emeritus and Distinguished Professor of Biology, is a virologist who received the Nobel Prize for his research into viral genetics. Baltimore was an organizer of the first Asilomar Conference on Recombinant DNA held in 1975 to discuss ethics and regulation of biotechnology. We sat down with him to discuss the debate over the origins of SARS-CoV-2.

What are the arguments that suggest SARS-CoV-2 is a naturally evolved virus? What is the evidence that suggests that it may have originated in and accidentally released from a laboratory in Wuhan, China?

The argument that it's a naturally evolved virus is from the belief that through the time of evolution, any sequence of RNA or DNA could evolve.

Biologists have seen what evolution can create: the whole natural world around us. We believe that evolution can do anything. But the fact that evolution might have been able to generate SARS-CoV-2 doesn't mean that that's how it came about. I think we very much need to find out what was happening in the Wuhan Institute of Virology. I think that we can't say for sure yet whether the SARS-CoV-2 virus came from natural origins or if it was genetically manipulated somehow.

Recently you were quoted as saying: "When I first saw the furin cleavage site in the viral sequence, with its arginine codons, I said to my wife it was the smoking gun for the origin of the virus. These features make a powerful challenge to the idea of a natural origin for SARS2." Can you unpack this quote for us?

Let me be clear, even though I used the phrase "smoking gun," I don't really think there's a smoking gun in the genome itself.

Now, within the SARS-CoV-2 genome there is an insertion of 12 nucleotides that is entirely foreign to the beta-coronavirus class of virus that SARS-CoV-2 is in. There are many other viruses in this class, including the closest relative of SARS-CoV-2 by sequence, and none of them have this sequence. The sequence is called the furin cleavage site.

To back up a little bit: In order to infect a cell, the spike protein on the surface of viruses like SARS-CoV-2 needs to first be cut, or cleaved. The cut needn't be terribly exact, but it needs to be cut. Different viruses attract different kinds of cellular "scissors," so to speak, to make this cut; the furin cleavage site attracts the furin protein providing the most efficient way to make a cut. You don't need a furin cleavage site to cut the protein, but it makes the virus more efficiently infectious.

So where did it come from in SARS-CoV-2? There are other viruses that have furin cleavage sites, other coronaviruses, though not the family of beta-coronaviruses. So this sequence's nucleotides could have hopped from some other virus. No one has identified a virus that has exactly this sequence, but it could have come from something close, then evolved into the sequence that we see today.

I'm perfectly willing to believe that happened, but I don't think it's the only way that that sequence could have appeared. The other way is that somebody could have put it in there. You can't distinguish between the two origins from just looking at the sequence. So, naturally, you want to know were there people in the virology laboratory in Wuhan who were manipulating viral genetic sequences? It's really a question of history: What happened?

When I first saw the sequence of the furin cleavage site—as I've said, other beta coronaviruses don't have that site—it seemed to me a reasonable hypothesis that somebody had put it in there. Now, I don't know if that's true or not, but I do know that it's a hypothesis that must be taken seriously.

Possible scenarios of virus evolution are discussed, with particular emphasis on the hypothesis that the virus could have emerged unintentionally through routine culture or gain-of-function experiments in a laboratory, where it was optimally adapted to human cells and caused cryptic infections among workers who finally spread the virus causing the pandemic.

It is important to note that all SARS-like coronaviruses known so far do not harbor this insert, while MERS-CoV, which docks to the DPP4 receptor, has an insert in this position of the protein consisting of four amino acids (Pro-Arg-Ser-Val) (Figure 2C). Furin cleavage sites are also found in other viruses in attachment proteins, including HIV, where the protease plays a role in entering the cell. However, a close similarity was found with the sequence of MERS-CoV. In Figure 2D, the nucleotide sequence around the PCS is compared between SARS-CoV-2 and MERS-CoV. Interstingly, on position 678 threonine is encoded by the same triplet ACT in SARS-CoV-2 and MERS-CoV, and on position 681 prolin is encoded again by the same triplet CCT and on position 682 arginine by CGG and CGC. Thus, there is not only a strong identity on the amino acid level, but also on the nucleotide level between SARS-CoV-2 and MERS-CoV. Given the code redundancy, the probability for an identical sequence encoding threonine (pos 678; which can be encoded by ACA, ACG, ACC and ACT) is 0.25×0.25=0.0625, and that of proline (pos 681; encoded by CCT, CCC, CCA and CCG) is again 0.0625. The overall probability harboring the same nucleotide sequence in these two positions is 0.0039. In position 682 with arginine coded by the sequence CGG (SARS-CoV-2) and CGC (MERS-CoV) we are again faced with a coincidence (nucleotides CG) of low probability [arginine is en coded by the codons CG (G,C,A,T), AGG, AGA]. In conclusion, there is a remarkable identity on amino acid and nucleotide level in and around the PCS between SARS-CoV-2 and MERS-CoV. This supports the hypothesis that the PCS/furin cleavage site was gained by a recombination event(s) involving these virus sequences. This notion is important in considering possible zoonotic events, placing laboratory events in the realm of the highly possible.

In this context, it is important to note that a sequence comparison of SARS-CoV-2 with other viruses revealed a 117-nucleotide sequence in the virus genome that is 94.6% identical to a human intron sequence of the netrin G1 gene. Several other viruses also contain human sequences, but they are much shorter (e.g., SARS-CoV contains a 41-nucleotide sequence). MERS-CoV does not contain a human sequence (40). The presence of a human sequence in SARS-CoV-2 supports the hypothesis that the progenitor was propagated in human cells where it gained the sequence by a recombination event...

Study finds virus was 'highly human adapted'

Scientists using computer modelling to study SARS-CoV-2, the virus that caused the COVID-19 pandemic, have discovered the virus is most ideally adapted to infect human cells - rather than bat or pangolin cells, again raising questions of its origin.

In a paper published in the Nature journal Scientific Reports, Australian scientists describe how they used high-performance computer modelling of the form of the SARS-CoV-2 virus at the beginning of the pandemic to predict its ability to infect humans and a range of 12 domestic and exotic animals.

Their work aimed to help identify any intermediate animal vector that may have played a role in transmitting a bat virus to humans, and to understand any risk posed by the susceptibilities of companion animals such as cats and dogs, and commercial animals like cows, sheep, pigs and horses.

The scientists, from Flinders University and La Trobe University, used genomic data from the 12 animal species to painstakingly build computer models of the key ACE2 protein receptors for each species. These models were then used to calculate the strength of binding of the SARS-CoV-2 spike protein to each species' ACE2 receptor.

Surprisingly, the results showed that SARS-CoV-2 bound to ACE2 on human cells more tightly than any of the tested animal species, including bats and pangolins. If one of the animal species tested was the origin, it would normally be expected to show the highest binding to the virus.

"Humans showed the strongest spike binding, consistent with the high susceptibility to the virus, but very surprising if an animal was the initial source of the infection in humans," says La Trobe University Professor David Winkler.

The findings, originally released on the ArXiv preprint server, have now been peer reviewed and published in Scientific Reports (Springer Nature).

"The computer modelling found the virus's ability to bind to the bat ACE2 protein was poor relative to its ability to bind human cells. This argues against the virus being transmitted directly from bats to humans. Hence, if the virus has a natural source, it could only have come to humans via an intermediary species which has yet to be found," says Flinders affiliated Professor Nikolai Petrovsky.

The team's computer modelling shows the SARS-CoV-2 virus also bound relatively strongly to ACE2 from pangolins, a rare exotic ant-eater found in some parts of South-East Asia with occasional instances of use as food or traditional medicines. Professor Winkler says pangolins showed the highest spike binding energy of all the animals the study looked at - significantly higher than bats, monkeys and snakes.

"While it was incorrectly suggested early in the pandemic by some scientists that they had found SARS-CoV-2 in pangolins, this was due to a misunderstanding and this claim was rapidly retracted as the pangolin coronavirus they described had less than 90% genetic similarity to SARS-CoV-2 and hence could not be its ancestor," Professor Petrovsky says.

This study and others have shown, however, that the specific part of the pangolin coronavirus spike protein that binds ACE2 was almost identical to that of the SARS-CoV-2 spike protein.

"This sharing of the almost identical spike protein almost certainly explains why SARS-CoV-2 binds so well to pangolin ACE2. Pangolin and SARS-CoV-2 spike proteins may have evolved similarities through a process of convergent evolution, genetic recombination between viruses, or through genetic engineering, with no current way to distinguish between these possibilities," Professor Petrovsky says.

"Overall, putting aside the intriguing pangolin ACE2 results, our study showed that the COVID-19 virus was very well adapted to infect humans."

"We also deduced that some domesticated animals like cats, dogs and cows are likely to be susceptible to SARS-CoV-2 infection too," Professor Winkler adds.

The extremely important and open question of how the virus came to infect humans has two main explanations currently. The virus may have passed to humans from bats through an intermediary animal yet to be found (zoonotic origin), but it cannot yet be excluded that it was released accidently from a virology lab. A thorough scientific, evidence-based investigation is needed to determine which of these explanations is correct.

How and where the SARS-CoV-2 virus adapted to become such an effective human pathogen remains a mystery, the researchers conclude, adding that finding the origins of the disease will help efforts to protect humanity against future coronavirus pandemics.

Australian scientist, the sole foreign researcher at the Wuhan lab, speaks out





It’s not that it’s impossible the virus spilled from there. Anderson, better than most people, understands how a pathogen can escape from a laboratory. SARS, an earlier coronavirus that emerged in Asia in 2002 and killed more than 700 people, subsequently made its way out of secure facilities a handful of times, she says.
If presented with evidence that such an accident spawned COVID-19, Anderson “could foresee how things could maybe happen,” she says. “I’m not naive enough to say I absolutely write this off.”




And yet, she still believes it most likely came from a natural source. Since it took researchers almost a decade to pin down where in nature the SARS pathogen emerged, Anderson says she’s not surprised they haven’t found the “smoking gun” bat responsible for the latest outbreak yet.
The Wuhan Institute of Virology is large enough that Anderson says she didn’t know what everyone was working on at the end of 2019. She is aware of published research from the lab that involved testing viral components for their propensity to infect human cells. Anderson is convinced no virus was made intentionally to infect people and deliberately released – one of the more disturbing theories to have emerged about the pandemic’s origins.



Despite this, Anderson does think an investigation is needed to nail down the virus’s origin once and for all. She’s dumbfounded by the portrayal of the lab by some media outside China, and the toxic attacks on scientists that have ensued.




One of a dozen experts appointed to an international taskforce in November to study the origins of the virus, Anderson hasn’t sought public attention, especially since being targeted by US extremists in early 2020 after she exposed false information about the pandemic posted online. The vitriol that ensued prompted her to file a police report. The threats of violence many coronavirus scientists have experienced over the past 18 months have made them hesitant to speak out because of the risk that their words will be misconstrued.
The elements known to trigger infectious outbreaks – the mixing of humans and animals, especially wildlife – were present in Wuhan, creating an environment conducive for the spillover of a new zoonotic disease. In that respect, the emergence of COVID-19 follows a familiar pattern. What’s shocking to Anderson is the way it unfurled into a global contagion.

The Wuhan Institute of Virology kept live bats in cages, new footage from inside the facility has revealed, disproving denials from World Health Organisation investigators who claimed the suggestion was a “conspiracy”.

An official Chinese Academy of Sciences video to mark the launch of the new biosafety level 4 laboratory in May 2017 speaks about the security precautions that are in place if “an accident” occurs and reveals there had been “intense clashes” with the French Government during the construction of the laboratory.

The video shows bats being held in a cage at the Wuhan Institute of Virology, along with vision of a scientist feeding a bat with a worm.

The 10 minute video is titled “The construction and research team of Wuhan P4 laboratory of Wuhan Institute of Virology, Chinese Academy of Sciences” and features interviews with its leading scientists.

The World Health Organisation report investigating the origin of the pandemic failed to mention that any bats had been kept at the Wuhan Institute of Virology and only its annex referred to animals being housed there. “The animal room in the P4 facility can handle a variety of species, including primate work with SARS-CoV-2,” it states.

A member of the World Health Organisation team investigating the origin of the pandemic in Wuhan, zoologist Peter Daszak said it was a conspiracy to suggest bats were held at the Wuhan Institute of Virology. In one tweet dated December, 2020 he said: “No BATS were sent to Wuhan lab for genetic analysis of viruses collected in the field. That’s now how this science works. We collect bat samples, send them to the lab. We RELEASE bats where we catch them!”

In another tweet, dated December 11, 2020, he said: “This is a widely circulated conspiracy theory. This piece describes work I’m the lead on and labs I’ve collaborated with for 15 years. They DO NOT have live or dead bats in them. There is no evidence anywhere that this happened. It’s an error I hope will be corrected.”

This month, Daszak appeared to retract his earlier denials and admitted the Wuhan Institute of Virology may have housed bats but admitted he had not asked them.

The Chinese Academy of Sciences video was discovered by researchers investigating the origin of the pandemic who call themselves DRASTIC. Digital archivist “Jesse” found the Chinese Academy of Sciences Video while the group's co-ordinator, who goes by a pseudonym of “Billy Bostickson” for safety reasons, has long complained evidence bats were housed in the Wuhan laboratories.

The video forms part of the investigation for the book “What Really Happened in Wuhan” which is available for pre-order at Amazon and Booktopia.

Mr Daszak has not responded to requests for comment.

The Case Against the Covid-19 Lab Leak Theory
Most theories about the pandemic starting with a bioengineered virus are less plausible than the simpler alternative: bats being bats.

In the past month, we’ve seen a second wave of interest in the theory that the Covid-19 pandemic began with a lab leak. Last spring, the media accurately reported the scientific consensus that Covid (also known as SARS CoV-2) is a natural virus that probably evolved much the same way as the last two deadly human coronaviruses, Severe Acute Respiratory Syndrome (SARS-COV) and Middle East Respiratory Syndrome (MERS-COV): namely, from bats, probably by way of an intermediate species like a raccoon dog, a ferret badger, or even a feral cat. Ever since the SARS outbreak of 2002-2003, after all, paper after paper and countless popular pieces have warned that sooner or later, nature would produce the next big SARS.

Now, some—latching onto new reporting, researchers’ open letter to Science magazine calling for greater investigation into the virus’s origins, and the Biden administration’s willingness to use the possibility of a lab leak to demand more transparency from China—argue media outlets were too quick to dismiss the lab leak theory. Substacker Matt Yglesias even called the media’s deeply skeptical coverage of lab leak theories a “fiasco” and “a huge fuckup.”

Despite the allure of this contrarianism, though, twenty years of post-SARS research into the origins and spread of bat coronaviruses point to a natural origin for Covid-19. Upon closer inspection, the so-called “new” evidence that has entranced pundits is neither new nor compelling. Inside the risky bat-virus engineering that links America to Wuhan

China emulated US techniques to construct novel coronaviruses in unsafe conditions.

In 2013, the American virologist Ralph Baric approached Zhengli Shi at a meeting. Baric was a top expert in coronaviruses, with hundreds of papers to his credit, and Shi, along with her team at the Wuhan Institute of Virology, had been discovering them by the fistful in bat caves. In one sample of bat guano, Shi had detected the genome of a new virus, called SHC014, that was one of the two closest relatives to the original SARS virus, but her team had not been able to culture it in the lab.

Baric had developed a way around that problem—a technique for “reverse genetics” in coronaviruses. Not only did it allow him to bring an actual virus to life from its genetic code, but he could mix and match parts of multiple viruses. He wanted to take the “spike” gene from SHC014 and move it into a genetic copy of the SARS virus he already had in his lab. The spike molecule is what lets a coronavirus open a cell and get inside it. The resulting chimera would demonstrate whether the spike of SHC014 would attach to human cells.

If it could, then it could help him with his long-term project of developing universal drugs and vaccines against the full spectrum of SARS-like viruses that he increasingly considered sources of potential pandemics. A SARS vaccine had been developed, but it wasn’t expected to be very effective against related coronaviruses, just as flu shots rarely work against new strains. To develop a universal vaccine that will elicit an antibody response against a gamut of SARS-like viruses, you need to show the immune system a cocktail of spikes. SHC014 could be one of them.

Baric asked Shi if he could have the genetic data for SHC014. “She was gracious enough to send us those sequences almost immediately,” he says. His team introduced the virus modified with that code into mice and into a petri dish of human airway cells. Sure enough, the chimera exhibited “robust replication” in the human cells—evidence that nature was full of coronaviruses ready to leap directly to people.

His 2015 paper, “A SARS-like cluster of circulating bat coronaviruses shows potential for human emergence,” was a tour de force, utilizing bleeding-edge genetic technology to alert the civilized world to a looming danger on its periphery. It also revived concerns about gain-of-function experiments, which Baric had known it would. In the paper, he spelled out the extra precautions he’d taken and held up the research as a test case. “The potential to prepare for and mitigate future outbreaks must be weighed against the risk of creating more dangerous pathogens,” he wrote. “Scientific review panels may deem similar studies building chimeric viruses based on circulating strains too risky to pursue.”

The NIH decided the risk was worth it. In a potentially fateful decision, it funded work similar to Baric’s at the Wuhan Institute of Virology, which soon used its own reverse-genetics technology to make numerous coronavirus chimeras.

Unnoticed by most, however, was a key difference that significantly shifted the risk calculation. The Chinese work was carried out at biosafety level 2 (BSL-2), a much lower tier than Baric’s BSL-3+.


What caused the covid-19 pandemic remains uncertain, and Shi says her lab never encountered the SARS-CoV-2 virus before the Wuhan outbreak. But now that US officials have said the possibility of a lab accident needs to be investigated, the spotlight has fallen on American funding of the Wuhan lab’s less safe research. Todaya chorus of scientists, including Baric, are coming forward to say this was a misstep. Even if there is no link to covid-19, allowing work on potentially dangerous bat viruses at BSL-2 is “an actual scandal,” says Michael Lin, a bioengineer at Stanford University.

The denial rests on the NIH’s specific definition of what was covered by the moratorium: work that would have deliberately enhanced SARS-like viruses, MERS, or flu by—for example—making them easier to spread through the air. The Chinese research did not have the specific goal of making the viruses more deadly, and rather than SARS itself, it used SARS’s close cousins, whose real-world risk to humans was unknown—in fact, determining the risk was the point of the research. Just as when you trade in part of a poker hand for fresh cards, there was no way of knowing whether the final chimeras would be stronger or weaker.

In 2014, the NIH awarded a five-year, $3.75 million grant to EcoHealth Alliance to study the risk that more bat-borne coronaviruses would emerge in China, using the same kind of techniques Baric had pioneered. Some of that work was to be subcontracted to the Wuhan Institute of Virology.

Two years later, Daszak and Shi published a paper reporting how the Chinese lab had engineered different versions of WIV1 and tested their infectiousness in human cells. The paper announced that the WIV had developed its own reverse-genetics system, following the Americans’ lead. It also included a troubling detail: the work, which was funded in part by the NIH grant, had been done in a BSL-2 lab. That meant the same viruses that Daszak was holding up as a clear and present danger to the world were being studied under conditions that, according to Richard Ebright, matched “the biosafety level of a US dentist’s office.”

Ebright believes one factor at play was the cost and inconvenience of working in high-containment conditions. The Chinese lab’s decision to work at BSL-2, he says, would have “effectively increas[ed] rates of progress, all else being equal, by a factor of 10 to 20”—a huge edge.

Work at the WIV was indeed progressing quickly. In 2017, Daszak and Shi followed with another study, also at BSL-2, that one-upped Baric’s work in North Carolina. The WIV had continued to unearth dozens of new SARS-like coronaviruses in bat caves, and it reported making chimeras with eight of them by fusing the spikes of the new viruses to the chassis of WIV1. Two of them replicated well in human cells. They were, for all intents and purposes, brand-new pathogens.

The revelation that the WIV was working with SARS-like viruses in subpar safety conditions has led some people to reassess the chance that SARS-CoV-2 could have emerged from some type of laboratory incident. “That’s screwed up,” the Columbia University virologist Ian Lipkin, who coauthored the seminal paper arguing that covid must have had a natural origin, told the journalist Donald McNeil Jr. “It shouldn’t have happened. People should not be looking at bat viruses in BSL-2 labs. My view has changed.”

But the WIV was not breaking any rules by working at BSL-2, says Filippa Lentzos, a biosecurity expert at King’s College London “There are no enforceable standards of what you should and shouldn’t do. It’s up to the individual countries, institutions, and scientists.” And in China, she says, the vertiginous rise of high-tech biological research has not been accompanied by an equivalent increase in oversight.

In an email, Zhengli Shi said she followed Chinese rules that are similar to those in the US. Safety requirements are based on what virus you are studying. Since bat viruses like WIV1 haven’t been confirmed to cause disease in human beings, her biosafety committee recommended BSL-2 for engineering them and testing them and BSL-3 for any animal experiments.

In response to questions about the decision to do the research in BSL-2 conditions, Peter Daszak forwarded a statement from EcoHealth Alliance stating that the organization “must follow the local laws of the countries in which we work” and that the NIH had determined the research was “not gain-of-function.”

There is no law against using tighter lab security, however, and according to Baric, these viruses deserve it. “I would never argue that WIV1 or SHC014 should be studied at BSL-2, because they can grow in primary human cells,” he says. “There’s some risk associated with those viruses. We have no idea whether they could actually cause severe disease in a human, but you want to err on the side of caution ... If you study a hundred different bat viruses, your luck may run out.”

Since the pandemic began, Baric has not said much about the possible origins of the virus or about his Chinese counterparts. On several occasions, however, he has quietly pointed to safety concerns at the WIV. In May 2020, when few scientists were willing to consider a lab leak in public, he published a paper acknowledging that “speculation about accidental laboratory escape will likely persist, given the large collections of bat virome samples stored in labs in the Wuhan Institute of Virology, the facility’s proximity to the early outbreak, and the operating procedures at the facility.” He flagged Daszak and Shi’s BSL-2 paper, in case anyone didn’t understand what he was saying.

In March, Dr. Shi Zhengli, the Wuhan-based virologist who has been accused of conducting risky experiments with bat coronaviruses, flatly denied allegations that the Wuhan Institute of Virology conducted studies with the military, an NBC News report said.

But the network reported that it uncovered evidence linking Shi to military scientists. She collaborated with Ton Yigang, a military scientist, on coronavirus research in 2018 and then with Zhou Yusen, another military scientist in December 2019. Zhou who was listed as deceased in the footnote of an article published in 2020, the report said. The report said it could not confirm the cause of his death.

David Asher, a former State Department adviser who co-authored a fact sheet last January on activity inside the lab, told NBC News that he is "confident" that the Chinese military was funding a "secret program" that involved coronaviruses.

Why won’t The Lancet admit it was wrong?

The journal is doubling down on its rejection of the lab leak hypothesis


Someone needs to tell The Lancet the most basic tenet of crisis management: when in a hole, stop digging. Instead, this world-renowned medical journal seems determined to keep shredding its reputation, in tandem with a group of experts ignorant to the damage they have caused the scientific community as they have stifled debate regarding the pandemic’s origins.

Early last year, just as the world was starting to grapple with the trauma of Covid, The Lancet published a highly-controversial statement in support of Chinese scientists, attacking “conspiracy theories suggesting that Covid-19 does not have a natural origin” and praising Beijing’s “rapid, open and transparent sharing of data”.

Clearly this was absurd given China’s cover-up, silencing of doctors and deletion of key data. But the statement, signed by 27 prominent experts, played a key role in shutting down suggestions the pandemic might have started with a lab incident, rather than spilling over naturally from animals. Scandalously, we later learned it was covertly drafted by British scientist Peter Daszak, £300,000-a-year president of Eco-Health Alliance charity and long-term partner of researchers at Wuhan Institute of Virology.

Despite the global furore, Daszak’s gang has gone back into battle with a follow-up statement, as I revealed they were planning to do 10 days ago. Even the headline on The Lancet article — Science, not speculation, is essential to determine how SARS-CoV-2 reached humans — seems designed to gaslight their critics, given their previous stance.

This latest statement is more nuanced but again disingenuous. It claims “the strongest clue” is that the virus evolved in nature, while saying suggestions of a lab leak “remain without scientifically validated evidence”. Yet there is zero firm evidence for natural spillover, and significant circumstantial evidence to raise suspicions of a lab incident. Besides, any leak could have involved a virus sampled from nature.

The theory that SARS-CoV-2 may have originated in a lab was considered a debunked conspiracy theory, but some experts are revisiting it amid calls for a new, more thorough investigation. Paul Thacker explains the dramatic U turn and the role of contemporary science journalism

For most of 2020, the notion that SARS-CoV-2 may have originated in a lab in Wuhan, China, was treated as a thoroughly debunked conspiracy theory. Only conservative news media sympathetic to President Donald Trump and a few lonely reports dared suggest otherwise. But that all changed in the early months of 2021, and today most outlets across the political spectrum agree: the “lab leak” scenario deserves serious investigation.

Understanding this dramatic U turn on arguably the most important question for preventing a future pandemic, and why it took nearly a year to happen, involves understanding contemporary science journalism.

A conspiracy to label critics as conspiracy theorists


Scientists and reporters contacted by The BMJ say that objective consideration of covid-19’s origins went awry early in the pandemic, as researchers who were funded to study viruses with pandemic potential launched a campaign labelling the lab leak hypothesis as a “conspiracy theory.”

A leader in this campaign has been Peter Daszak, president of EcoHealth Alliance, a non-profit organisation given millions of dollars in grants by the US federal government to research viruses for pandemic preparedness.1 Over the years EcoHealth Alliance has subcontracted out its federally supported research to various scientists and groups, including around $600 000 (£434 000; €504 000) to the Wuhan Institute of Virology.1

Shortly after the pandemic began, Daszak effectively silenced debate over the possibility of a lab leak with a February 2020 statement in the Lancet.2 “We stand together to strongly condemn conspiracy theories suggesting that covid-19 does not have a natural origin,” said the letter, which listed Daszak as one of 27 coauthors. Daszak did not respond to repeated requests for comment from The BMJ.

“It’s become a label you pin on something you don’t agree with,” says Nicholas Wade, a science writer who has worked at Nature, Science, and the New York Times. “It’s ridiculous, because the lab escape scenario invokes an accident, which is the opposite of a conspiracy.”

But the effort to brand serious consideration of a lab leak a “conspiracy theory” only ramped up. Filippa Lentzos, codirector of the Centre for Science and Security Studies at King’s College, London, told the Wall Street Journal, “Some of the scientists in this area very quickly closed ranks.”3 She added, “There were people that did not talk about this, because they feared for their careers. They feared for their grants.”

Daszak had support. After he wrote an essay for the Guardian in June 2020 attacking the former head of MI6 for saying that the pandemic could have “started as an accident,” Jeremy Farrar, director of the Wellcome Trust and co-signer of the Lancet letter, promoted Daszak’s essay on Twitter, saying that Daszak was “always worth reading.”4

Daszak’s behind-the-scenes role in orchestrating the statement in the Lancet came to light in November 2020 in emails obtained through freedom of information requests by the watchdog group US Right To Know.

“Please note that this statement will not have EcoHealth Alliance logo on it and will not be identifiable as coming from any one organization or person,” wrote Daszak in a February email, while sending around a draft of the statement for signatories.5 In another email, Daszak considered removing his name from the statement “so it has some distance from us and therefore doesn’t work in a counterproductive way.”6

Several of the 27 scientists who signed the letter Daszak circulated did so using other professional affiliations and omitted reporting their ties to EcoHealth Alliance.3

For Richard Ebright, professor of molecular biology at Rutgers University in New Jersey and a biosafety expert, scientific journals were complicit in helping to shout down any mention of a lab leak. “That means Nature, Science, and the Lancet,” he says. In recent months he and dozens of academics have signed several open letters rejecting conspiracy theory accusations and calling for an open investigation of the pandemic’s origins.789

“It’s very clear at this time that the term ‘conspiracy theory’ is a useful term for defaming an idea you disagree with,” says Ebright, referring to scientists and journalists who have wielded the term. “They have been successful until recently in selling that narrative to many in the media.”

The Lancet’s editor in chief, Richard Horton, did not respond to repeated requests for comment but, after The BMJ had sent him questions, the Lancet expanded Daszak’s conflicts of interest on the February statement and recused him from working on its task force looking into the pandemic’s origin.1011

The Lancet letter ultimately helped to guide almost a year of reporting, as journalists helped to amplify Daszak’s message and to silence scientific and public debate. “We’re in the midst of the social media misinformation age, and these rumours and conspiracy theories have real consequences,” Daszak told Science.12 Months later in Nature, he again criticised “conspiracies” that the virus could have come from the Wuhan Institute of Virology and complained about “politically motivated organisations” requesting his emails.13

That summer Scientific American, one of the oldest and best known popular science magazines in America, published a complimentary profile of Daszak’s colleague, Shi Zhengli, a centre director at the Wuhan Institute of Virology, which has been funded by EcoHealth Alliance.14

EcoHealth Alliance and the Wuhan Institute of Virology earned additional sympathetic reporting after the US National Institutes of Health (NIH) cancelled its grant to EcoHealth Alliance in April last year—allegedly on President Trump’s order—because of its ties to Wuhan, a decision protested by 77 Nobel laureates and 31 scientific societies.15 (The NIH has subsequently awarded EcoHealth Alliance new funding.)

Efforts to characterise the lab leak scenario as unworthy of serious consideration were far reaching, sometimes affecting reporting that had first appeared well before the covid-19 pandemic. For example, in March 2020 Nature Medicine added an editor’s note (“Scientists believe that an animal is the most likely source of the coronavirus”) to a 2015 paper on the creation of a hybrid version of a SARS virus, co-written by Shi.16

Wade explains, “Science journalists differ a lot from other journalists in that they are far less sceptical of their sources and they see their main role as simply to explain science to the public.” This, he says, is why they began marching in unison behind Daszak.


The U turn


By the end of 2020, just a handful of journalists had dared to seriously discuss the possibility of a lab leak. In September, Boston magazine reported on a preprint that found the virus unlikely to have come from the Wuhan seafood market, as Daszak has argued, and that it seemed too well adapted to humans to have arisen naturally. However, the story failed to garner much attention, similarly to a little noticed investigative report by the Associated Press in December that exposed how the Chinese government was clamping down on research into covid-19’s origins.

In January this year, New York magazine ran a sprawling story detailing how the pandemic could have started with a leak from the lab in Wuhan. The hypothetical scenario: “SARS-CoV-2, the virus that causes covid-19, began its existence inside a bat, then it learned how to infect people in a claustrophobic mine shaft, and then it was made more infectious in one or more laboratories, perhaps as part of a scientist’s well-intentioned but risky effort to create a broad-spectrum vaccine.” Scientists and their media allies swiftly criticised the article.

But mainstream outlets from the New York Times to the Washington Post are now treating the lab leak hypothesis as a worthy question, one to be answered with a serious investigation. In a recent interview with the New York Times, Shi denied that her lab was ever involved in “gain of function” experiments (box 1) that enhance a virus’s virulence. But the newspaper reported that her lab had been involved in experiments that altered the transmissibility of viruses, alongside interviews with scientists who said that far more transparency was necessary to determine the truth of SARS-CoV-2’s origins.17

Two major events are probably responsible for the media’s change in tune. First, Trump was no longer president. Because Trump had said that the virus could have come from a Wuhan lab, Daszak and others used him as a convenient foil to attack their critics. But the framing of the lab leak hypothesis as a partisan issue was harder to sustain after Trump left the White House.

Second, after months of negotiation the Chinese government finally allowed the World Health Organization to come to Wuhan and investigate the pandemic’s origin. But in January 2021 WHO, which included Daszak on the team, returned with no evidence that the virus had arisen through natural spill-over.18 More worryingly, members were allowed only a few hours of supervised access to the Wuhan Institute of Virology.

The White House then released a statement making clear that it did not trust China’s propaganda denying that the virus could have come from one of the country’s labs. “We have deep concerns about the way in which the early findings of the covid-19 investigation were communicated and questions about the process used to reach them,” said the statement. “It is imperative that this report be independent, with expert findings free from intervention or alteration by the Chinese government.”

The following month the Washington Post editorial board called for an open and transparent investigation of the virus’s origins, highlighting Shi’s experiments with bat coronaviruses that were genetically very similar to the one that caused the pandemic.19 It asked, “Could a worker have gotten infected or inadvertent leakage have touched off the outbreak in Wuhan?” The Wall Street Journal, citing a US intelligence document, recently reported that three Wuhan Institute of Virology researchers were admitted to hospital in November 2019.20

To follow any US financial ties and to better understand how the pandemic started, Republicans have launched investigations of government agencies that fund coronavirus research, and one investigative committee has sent a letter to Daszak at EcoHealth Alliance demanding that he turn over documents. Meanwhile, Senate Republicans and Democrats have started to discuss an independent investigation of the virus’s origins.

A hard truth to swallow


The growing tendency to treat the lab leak scenario as worthy of serious investigation has put some reporters on the defensive. After Robert Redfield, former director of the Centers for Disease Control and Prevention, appeared on CNN in March, Scientific American’s editor in chief, Laura Helmuth, tweeted, “On CNN, former CDC director Robert Redfield shared the conspiracy theory that the virus came from the Wuhan lab.” The following day, Scientific American ran an essay calling the lab leak theory “evidence free.” And a week later a Nature reporter, Amy Maxmen, labelled the idea that the virus could have leaked from a lab as “conjecture.”

Helmuth did not respond to questions from The BMJ.

Some media outlets have attempted to justify their past reporting about the lab leak hypothesis as simply a matter of tracking a “scientific consensus” which, they say, has now changed. Vox posted an erratum noting, “Since this piece was originally published in March 2020, scientific consensus has shifted.”

The “scientific consensus” argument does not sit well with David Relman, a microbiologist at Stanford University, California. “We can’t even begin to talk about a consensus other than a consensus that we don’t know [the origins of SARS-CoV-2],” he recently told the Washington Post.21

A year lost


While the narrative took months to change in the media, several high profile intelligence sources had treated the lab leak theory seriously from early on. In April 2020, Avril Haines joined two other former deputy directors of the Central Intelligence Agency to write an essay in Foreign Policy asking, “To what extent did the Chinese government misrepresent the scope and scale of the epidemic?”22 A week later, one of the former intelligence officials who wrote that essay gave similar quotes to Politico.

Ignoring these early warnings led to a year of biased, failed reporting, says Wade. “They didn’t question what their sources were saying,” he says of the reporters who helped to sell the conspiracy theory narrative to the public. “That is the simple explanation for this phenomenon.”

An impartial, credible investigation?


As the news media scramble to correct and reflect on what went wrong with nearly a year of reporting, the episode has also highlighted quality control issues at the ubiquitous “fact checking” services.

Prominent outlets such as PolitiFact23 and FactCheck.org24 have added editor’s notes to pieces that previously “debunked” the idea that the virus was created in a lab or could have been bioengineered—softening their position to one of an open question that is “in dispute.” For almost a year Facebook sought to control misinformation by banning stories suggesting that the coronavirus was man made. After renewed interest in the virus’s origin, Facebook lifted the ban.25

Whether a credible investigation will be made into the lab leak scenario remains to be seen. WHO and the Lancet both launched investigations last year (box 2), but Daszak was involved in both, and neither has made significant progress.

In recent weeks, several high profile scientists who once denigrated the idea that the virus could have come from a lab have made small steps into demanding an open investigation of the pandemic’s origin.

The NIH’s director, Francis Collins, said in a recent interview, “The Chinese government should be on notice that we have to have answers to questions that have not been answered about those people who got sick in November who worked in the lab and about those lab notebooks that have not been examined.” He added, “If they really want to be exonerated from this claim of culpability, then they have got to be transparent.”26

But the nature of this investigation has still not been decided.

A Group of Scientists Presses a Case Against the Lab Leak Theory of Covid

In the latest volley of the debate over the origins of the coronavirus, a group of scientists this week presented a review of scientific findings that they argue shows a natural spillover from animal to human is a far more likely cause of the pandemic than a laboratory incident.

Among other things, the scientists point to a recent report showing that markets in Wuhan, China, had sold live animals susceptible to the virus, including palm civets and raccoon dogs, in the two years before the pandemic began. They observed the striking similarity that Covid-19’s emergence had to other viral diseases that arose through natural spillovers, and pointed to a variety of newly discovered viruses in animals that are closely related to the one that caused the new pandemic.

GENEVA, June 10 (Xinhua) -- The lab-leak theory that the COVID-19 virus escaped from a Chinese laboratory is "extremely unlikely," a renowned German virologist has told Swiss digital magazine Republik.

Asked about the origins of the coronavirus and the theory that it accidentally escaped from the Wuhan Institute of Virology, Christian Drosten, a German virologist at the Berlin-based Charite Institute of Virology, said "this idea of a research accident is extremely unlikely for me because it would be far too cumbersome."

Drosten, who has been researching coronaviruses for years and also played a crucial role in the discovery of the SARS-associated coronavirus in 2003, said the most likely origin was carnivore breeding and the fur industry.

In the article "Mr. Drosten, where did this virus come from?," Drosten told the Republik journalists that he has no evidence for the conjecture, except for the clearly proven origin of SARS-CoV-1, a virus of the same species.

"Viruses of the same species do the same things and often come from the same source," he was quoted as saying, noting that the scientifically documented transitional hosts of the SARS virus were raccoon dogs and crawling cats...

But Richard Ebright, a molecular biologist at Rutgers University and a persistent critic of attempts to diminish the likelihood of a laboratory leak, said that this was a straw-man argument.

Dr. Ebright said it was possible that a W.I.V. lab worker might have contracted the coronavirus on a field expedition to study bats or while processing a virus at the lab. The new paper, he argued, failed to address such possibilities.

“The review does not advance the discussion,” Dr. Ebright said.
