      WASHINGTON (SBG) - A classified study of the origin of SARS-CoV-2 conducted a year ago by scientists at the Lawrence Livermore National Laboratory, the Department of Energy?s premier biodefense research institution, concluded the novel coronavirus at the heart of the current pandemic may have originated in a laboratory in China, Sinclair has learned.

      Researchers at Livermore?s ?Z Division,? the lab?s intelligence unit, issued the report May 27, 2020, classified ?Top Secret.? Its existence is previously undisclosed. The Z Division report assessed that both the lab-origin theory and the zoonotic theory were plausible and warranted further investigation. Sinclair has not reviewed the report but confirmed its contents through interviews with multiple sources who read it or were briefed on its contents.

      In an email to Sinclair, a Livermore spokesperson confirmed the existence of the report but declined to provide additional information. ?Because the report you are referring to is classified,? wrote Lynda Seaver, director of public affairs, ?it would be inappropriate for our lab to discuss this.?

        We are currently in a rapidly expanding pandemic of the SARS-CoV-2 virus, which originated in the city of Wuhan in central China. The disease COVID-19 is now spread worldwide and has tremendous socio-economic consequences. The origin of the virus can be reconstructed through epidemiological studies and, even more so, from genome comparisons. How the evolution of the virus and the transition to humans might have happened is the subject of much speculation. It is considered certain that the virus is of animal origin and very likely passed from bats to humans in a zoonotic eve..nt. An intermediate host was postulated, but many SARS-like bat viruses have the ability to infect human cells directly, which has been shown experimentally by scientists in the Wuhan Institute of Virology using collected specimens containing virus material from horseshoe bats. The propagation of SARS-like bat viruses in cell culture allowed experiments aimed at increasing the infectivity of the virus and adaptation to human cells. This article summarizes the unique properties of SARS-CoV-2 and focusses on a specific sequence encoding the spike protein. Possible scenarios of virus evolution are discussed, with particular emphasis on the hypothesis that the virus could have emerged unintentionally through routine culture or gain-of-function experiments in a laboratory, where it was optimally adapted to human cells and caused cryptic infections among workers who finally spread the virus causing the pandemic.

        Summary and Conclusion


        The current COVID-19 pandemic caused by the SARS-CoV-2 coronavirus has already caused immeasurable suffering and economic damage, and its long-term socio-economic impact cannot yet be assessed. Never before has an event caused more death and long-term suffering, and the associated crisis management brought trade, traffic, travel, social activities and even family contacts to a standstill. Therefore, the question of how this pandemic came about and when the primary zoonotic event took place is important to answer in order to be able to prevent further zoonotic events of this kind.

        Although it was initially believed that the virus could not be transmitted from person to person, the opposite soon turned out. The intensity of the wave of infection is favored by the high infectivity of the virus, by symptom-free carriers and, in symptomatic cases, by a symptom-free period lasting several days. It is considered as certain that SARS-CoV-2 is of bat origin and that there must have been gain of function events through insertions and point mutations by which adaptation of the predecessor virus occurred to human cells equipped with the ACE2 receptor, with neuropilin-1 and the proteases furin and TMPRSS2, all of which are ideally suited for an optimal virus entry into human cells.

        SARS-CoV-2 has the greatest similarity in the nucleic acid and amino acid sequence to a virus (RaTG13) that has been isolated from swabs of bats, endemic in central China, at the Institute of Virology in Wuhan and propagated there in cell culture. Although critical sequences in the spike protein, in particular the insertion of a twelve-nucleotide section in the polybasic cleavage site, could have been acquired naturally, for example through recombination events with a SARS-like corona virus harboring the same sequence, it is highly unliklely that the selection occurred in bats or in an intermediate animal host. However, a scenario seems likely, according to which the selection for a highly infectious agent took place in human cells, notably in cell culture, as a byproduct of virus propagation and experimental work. Sequence comparison revealed that the insert creating the PCS/furine cleavage site is partially identical to an insert in the spike gene of MERS-CoV. Based on this, the hypothesis was proposed that during coinfection of human cells equipped with ACE2 and DPP4, RaTG13 (or a similar progenitor) gained the sequence from MERS-CoV (or a similar virus harboring the sequence) by recombination, through which it became better propagating in vitro, which is beneficial for the experimentation (selection for high virus titer). At the same time the infectivity of the virus was enhanced allowing unintentional, cryptic infections of employees, which was the starting point for the pandemic.

        Furthermore, the possibility that the progenitor virus was propagated in human ACE2 transgenic animals, which would accelerate human adaptation, should also be considered. At this point it should be remembered that releases of pathogenic viruses from the laboratory have already happened, which was a matter of serious concern in China years ago. Back in 2003, in Kunming (province Yunnan), a hantavirus outbreak occurred, and it was attributed to laboratory rats in which two hantavirus strains multiplied and generated a new type of virus through recombination. Students who had worked with the animals became infected with this. The authors of the report (published in 2010) concluded prophetically that ?This study sends a timely warning that laboratory exposure remains an important source of hantavirus infection in China and that new strains continue to emerge via reassortment and recombination of the RNA genome segments? (47). There is no reason to believe that the same cannot happen with other virus strains. The coincidence of the outbreak of the disease COVID-19 in Wuhan and intensive ongoing work with SARS-like viruses at a research institute located there, housing the largest corona virus bank in Asia, has also sparked the public discussion whether gain-of-function experiments contributed to the pandemic. Even though evidence is lacking that the virus was intentionally genetically engineered, the possibility that the virus could have emerged unintentionally through laboratory experiments should lead to a rethinking of the need for gain-of-function experiments aimed at enhancing the pathogenicity of a disease-causing agent.

          It is a shame to know that the wrongdoers in the United States and China are still covered with honors when they have produced such a mass of suffering, because the presidents of these two super powers cover them.

          This shame must be partly shared by all those scientists who knew and did not say anything for x or y reasons.

          These practices were prohibited in the USA, which is why one of the culprits financed their realization in China.

          But now, what can we do ?

          If these two presidents have no honor, nothing will change.

            Sir Richard Dearlove said aspects of the virus "point in the direction of it being somewhat tailored" though he warned this may never be proven.

            The former "C" of the Secret Intelligence Service ? equivalent to "M" in James Bond ? also told LBC's Tom Swarbrick that more information on the coronavirus' origin will soon come out.

            Some have theorised the coronavirus could have escaped from the Wuhan Institute of Virology. Work to establish the origin of the virus is ongoing.

            Sir Richard, who headed up the spy agency between 1999 and 2004, told Tom the World Health Organisation's report, which said a lab leak was highly unlikely but further work was needed, was a "farcical investigation"...

                ?One Health? approach against coronaviruses. Risk analysis for coronavirus infections might also consider the variability of the host genome whose level is different across breeds and populations, as it might be derived from their genetic histories.

                Before 2020, the rules followed by virologists in China and elsewhere required that experiments with the SARS1 and MERS viruses be conducted in BSL3 conditions. But all other bat coronaviruses could be studied in BSL2, the next level down. BSL2 requires taking fairly minimal safety precautions, such as wearing lab coats and gloves, not sucking up liquids in a pipette, and putting up biohazard warning signs. Yet a gain-of-function experiment conducted in BSL2 might produce an agent more infectious than either SARS1 or MERS. And if it did, then lab workers would stand a high chance of infection, especially if unvaccinated.

                Much of Dr. Shi?s work on gain-of-function in coronaviruses was performed at the BSL2 safety level, as is stated in her publications and other documents. She has said in an interview with Science magazine that ?The coronavirus research in our laboratory is conducted in BSL-2 or BSL-3 laboratories.?

                Concern about safety conditions at the Wuhan lab was not, it seems, misplaced. According to a fact sheet issued by the State Department on January 21,2021, ? The U.S. government has reason to believe that several researchers inside the WIV became sick in autumn 2019, before the first identified case of the outbreak, with symptoms consistent with both COVID-19 and common seasonal illnesses.?

                David Asher, a fellow of the Hudson Institute and former consultant to the State Department, provided more detail about the incident at a seminar. Knowledge of the incident came from a mix of public information and ?some high end information collected by our intelligence community,? he said. Three people working at a BSL3 lab at the institute fell sick within a week of each other with severe symptoms that required hospitalization. This was ?the first known cluster that we?re aware of, of victims of what we believe to be COVID-19.? Influenza could not completely be ruled out but seemed unlikely in the circumstances, he said.

                In the case of SARS1, researchers have documented the successive changes in its spike protein as the virus evolved step by step into a dangerous pathogen. After it had gotten from bats into civets, there were six further changes in its spike protein before it became a mild pathogen in people. After a further 14 changes, the virus was much better adapted to humans, and with a further 4 the epidemic took off.

                But when you look for the fingerprints of a similar transition in SARS2, a strange surprise awaits. The virus has changed hardly at all, at least until recently. From its very first appearance, it was well adapted to human cells. Researchers led by Alina Chan of the Broad Institute compared SARS2 with late stage SARS1, which by then was well adapted to human cells, and found that the two viruses were similarly well adapted. ?By the time SARS-CoV-2 was first detected in late 2019, it was already pre-adapted to human transmission to an extent similar to late epidemic SARS-CoV,? they wrote.

                Even those who think lab origin unlikely agree that SARS2 genomes are remarkably uniform. Dr. Baric writes that ?early strains identified in Wuhan, China, showed limited genetic diversity, which suggests that the virus may have been introduced from a single source.?

                A single source would of course be compatible with lab escape, less so with the massive variation and selection which is evolution?s hallmark way of doing business.

                The uniform structure of SARS2 genomes gives no hint of any passage through an intermediate animal host, and no such host has been identified in nature.

                Proponents of natural emergence suggest that SARS2 incubated in a yet-to-be found human population before gaining its special properties. Or that it jumped to a host animal outside China.

                All these conjectures are possible, but strained. Proponents of lab leak have a simpler explanation. SARS2 was adapted to human cells from the start because it was grown in humanized mice or in lab cultures of human cells, just as described in Dr. Daszak?s grant proposal. Its genome shows little diversity because the hallmark of lab cultures is uniformity.

                Leaked Chinese document reveals a sinister plan to ?unleash? coronaviruses

                A leaked 2015 government paper has revealed a discussion by top Chinese scientists who said a virus could be ?unleashed in way never seen before?.

                A document written by Chinese scientists and Chinese public health officials in 2015 discussed the weaponisation of SARS coronavirus, reveals the Weekend Australian.

                Titled The Unnatural Origin of SARS and New Species of Man-Made Viruses as Genetic Bioweapons, the paper predicted that World War Three would be fought with biological weapons.

                Released five years before the start of the COVID-19 pandemic, it describes SARS coronaviruses as a ?new era of genetic weapons? that can be ?artificially manipulated into an emerging human ?disease virus, then weaponised and unleashed in a way never seen before?.

                Peter Jennings, the executive director of the Australian Strategic Policy Institute (ASPI), told news.com.au that the document is as close to a ?smoking gun? as we?ve got.

                ?I think this is significant because it clearly shows that Chinese scientists were thinking about military application for different strains of the coronavirus and thinking about how it could be deployed,? said Mr Jennings.

                ?It begins to firm up the possibility that what we have here is the accidental release of a pathogen for military use,? added Mr Jennings.

                He also said that the document may explain why China has been so reluctant for outside investigations into the origins of COVID-19.

                ?If this was a case of transmission from a wet market it would be in China?s interest to co-operate ? we?ve had the opposite of that.?

                Among the 18 listed authors of the document are People?s Liberation Army scientists and weapons experts.

                Robert Potter, a cyber security specialist who analyses leaked Chinese government documents was asked by The Australian to verify the paper. He says the document definitely isn?t fake.

                  Dozens of think tanks, inteligence agencies and academic institutions have written papers about the dangers of weaponising a vast range of natural and artificial pathogens. There is a whole industry based on this possibility, principally in the US. That the Chinese are also concerning themselves with it is hardly a smoking gun. We have the tools to make anything we like what we do not have is the knowledge to know how the the thing we make will behave. Even the SARs pandemic virus we have been studying for the last year is throwing up mutations but we can not look at the changed genome and tell what effect it is going to have, we have to wait until it has infected lots of people and then see if it made any difference. This is a million miles away from taking flu, a CoV or anything else and redesigning its genome to be more or less lethal. To be a useful bio-weapon, for a nation state, they would also need a 100% effective vaccine for their own population and be sure it would not mutate. This is all science fiction at this point in time. A nutter who just wanted to reduce the global population is the real danger not nation states and they are far more likely just to use an existing hi-path human disease.

                If you hear anyone claim ?we know the virus didn?t come from a lab?, don?t buy it ? it may well have. Labs around the globe have been creating synthetic viruses like CoV2 for years. And no, its genome would not necessarily contain hallmarks of human manipulation: modern genetic engineering tools permit cutting and pasting genomic fragments without leaving a trace. It can be done quickly, too: it took a Swiss team less than a month to create a synthetic clone of CoV2.

                How I Learned to Start Worrying

                Oh, come on. Lab-made? Nonsense! Back in January, that was my knee-jerk reaction when ideas that Covid-19 is caused by a laboratory leak had just surfaced. Bioweapon? Well, that is just Flat Earth crazies territory. Thus, whenever I kept hearing anything about non-natural origins of SARS-CoV-2, I brushed it aside under similar sentiments. So what if there is a virology institute in Wuhan? Who knows how many of those are sprinkled throughout China.

                At some point, it became necessary to brush such theories aside in a substantiated manner, as their proponents began to back up their theses about the possible artificial nature of the virus with arguments from molecular biology, and when engaging them in debate, I wanted to smash their conspiracy theories with cold, hard scientific facts. Just like that Nature paper (or so I thought).

                So it was then, in pursuit of arguments against the virus?s lab-madeness, that I got infected by the virus of doubt. What was the source of my doubts? The fact that the deeper you dive into the research activities of coronavirologists over the past 15?20 years, the more you realize that creating chimeras like CoV2 was commonplace in their labs. And CoV2 is an obvious chimera (though not nesessarily a lab-made one), which is based on the ancestral bat strain RaTG13, in which the receptor binding motif (RBM) in its spike protein is replaced by the RBM from a pangolin strain, and in addition, a small but very special stretch of 4 amino acids is inserted, which creates a furin cleavage site that, as virologists have previously established, significantly expands the ?repertoire? of the virus in terms of whose cells it can penetrate. Most likely, it was thanks to this new furin site that the new mutant managed to jump species from its original host to humans...
