New filovirus found in bats in Spain

The red text is false. No new documented Ebola transmission with a 42-day incubation period has been found; the correct reference is that WHO and African authorities as a precautionary measure wait twice the maximum incubation period before declaring an outbreak contained. Indeed, no human Ebola or Marburg cases have been detected ANYWHERE in the world since January 1, 2009.

Besides this obvious error, the article seems to indicate that the bat deaths occurred several years ago, while the identification of the virus likely just occurred.

The ebola virus found in the remains of dead bats in Asturias most
likely belongs to a species [of ebolavirus] harmless to humans,
according to Francisco Llinares, Professor of Microbiology at the
University of San Pablo CEU. The professor referred to this case
during the lecture "Filoviridae: Ebola and Marburg," within the
framework of the IV Conference of Microbiology and Public Health at
the San Pablo University CEU, this year [2010] entitled "The coming Virus."

However, he said that "only the results of the laboratory analysis
will determine exactly what type of ebolavirus we found." Llinares
believes this is surely a case of a "new type" [of ebolavirus], i.e.
not as yet documented. In fact, he recalled scientific reports that,
years ago, unexplained deaths of bats had been observed in Asturias.
Only when by chance a group of researchers came to check [on these
bat deaths] did they realize that an ebolavirus was the cause of the mortality.

The above mentioned [laboratory] analysis to distinguish the type of
virus [responsible for the bat die-off] was carried out in France in
a P4 safety level lab, and [Llinares] stated that "Spain cannot
handle this type of virus, because the highest level of laboratory
safety in Spain is P3," he said.

Throughout his speech, Llinares also referred to the [apparent]
evolution of viruses in recent outbreaks that have been documented.
On this point, he expressed concern about the fact that in the recent
cases of [ebolavirus] infection in Uganda and the Congo, the
incubation period has been double what was previously measured. The
previously seen maximum incubation period of 21 days had changed to
42 days, a circumstance that "lengthens the time of possible
infection," and therefore "can help to [further disseminate the virus]."


In spite of the risk and the mortality that the virus carries,
Llinares said that "Ebolavirus is not the new rider of the
Apocalypse." As he noted, "outbreaks have occurred in a very specific
zone -- Equatorial Africa -- which is also very heavily under
surveillance by the World Health Organization."

With regard to possible antidotes, what science can say now is that
"there is no treatment or vaccine." On this point, he reviewed the
research to produce a vaccine that induces the production of
antibodies against the ebolavirus glycoprotein, a protein that the
[ebolavirus] uses to bind to the receptor cell membranes. He also
mentioned the recent discovery that a high percentage of the
population of Gabon has developed antibodies to [ebolavirus] without
having previously developed hemorrhagic fever.

[The RNA negative strand viruses belonging to the family
_Filoviridae_ are classified into 2 genera, one of which is
designated the genus _Marburgvirus_ and the other the genus
_Ebolavirus_. The basis for assigning the Asturias bat viruses
described above to the genus _Ebolavirus_ is unclear. Filoviruses
viruses exhibit a characteristic bacilliform morphology, which may be
the basis for the identification of these viruses rather than the
immunological and genetic characterisation necessary to place them in
the genus _Ebolavirus_.


Not all ebolaviruses are pathogenic for humans, which might explain
the cryptic presence of such viruses in Spain. Cote d'Ivoire
ebolavirus, for example, has been associated with only one case of
human disease, and the Reston ebolaviruses, although pathogenic for
cynomolgous monkeys and swine, are apathogenic in humans. Further
information on the characterisation and distribution of the Asturias
bat filoviruses would be welcomed.


Filoviruses have been frequently isolated from bats, and some species
have been implicated in the transmission of hemorrhagic disease to
humans. Information on the species classification and biology of the
virus-infected bats in Asturias would be welcomed also.

A novel filovirus has been detected in bats in the Iberian Peninsula. A
full length genome sequence has been obtained in a collaboration between
the Instituto de Salud Carlos III and the Center for Infection and Immunity
at Columbia University. Preliminary phylogenetic analysis indicates that
this virus is taxonomically distinct from ebolavirus and Lake Victoria
marburgvirus. There is no evidence that this virus has infected humans
.

[ProMED-mail is indebted to Antonio Tenorio for providing the above
information clarifying the status of the filovirus isolated from bats in
the north of Spain.

Currently, the family _Filoviridae_ consists of 2 distinct genera
_Ebolavirus_ and _Marburgvirus_ (Virus Taxonomy, 8th Report of the ICTV,
Elsevier, 2005), embracing viruses predominately associated with outbreaks
of haemorrhagic disease in the African continent. It is likely that this
novel virus found in bats in the north of Spain, and as yet not associated
with disease in humans, may have to be ranked by the International
Committee on Taxonomy of Viruses (ICTV) as a novel genus in the family
_Filoviridae_. Further characterization of the properties and host range of
this novel bat virus is awaited with great interest by the virology community.

Abstract

Filoviruses, amongst the most lethal of primate pathogens, have only been reported as natural infections in sub-Saharan Africa and the Philippines. Infections of bats with the ebolaviruses and marburgviruses do not appear to be associated with disease. Here we report identification in dead insectivorous bats of a genetically distinct filovirus, provisionally named Lloviu virus, after the site of detection, Cueva del Lloviu, in Spain.

First Ebola-like virus native to Europe discovered

New virus could be the first filovirus to cause disease in bats

A team of international researchers has discovered a new Ebola-like virus ? Lloviu virus -- in bats from northern Spain. Lloviu virus is the first known filovirus native to Europe, they report in a study published in the journal PLOS Pathogens on Octobr 20th.

The study was a collaboration among scientists at the Center for Infection and Immunity (CII) at Columbia University's Mailman School of Public Health, the Instituto de Salud Carlos III (ISCIII) in Spain, Roche Life Sciences, Centro de Investigaci?n Pr?ncipe Felipe, Grupo Asturiano para el Estudio y Conservaci?n de los Murci?lagos, Consejo Suerior de Investigaciones Cient?ficas and the Complutense University in Spain.

Filoviruses, which include well-known viruses like Ebola and Marburg, are among the deadliest pathogens in humans and non-human primates, and are generally found in East Africa and the Philippines. The findings thus expand the natural geographical distribution of filoviruses.

"The study is an opportunity to advance the knowledge of filoviruses' natural cycle," said Ana Negredo, one of the first authors of the study.

Scientists at ISCIII analyzed lung, liver, spleen, throat, brain and rectal samples from 34 bats found in caves in Asturias and Cantabria, Spain, following bat die-offs in France, Spain and Portugal in 2002 affecting mainly one bat species.

They screened these samples for a wide range of viruses using the polymerase chain reaction, a molecular technique that allows scientists to amplify genetic material, and. detected a filovirus. Filoviruses include ebolaviruses and marburgviruses, two viruses associated with severe disease in humans and other primates..

CII scientists used high-throughput sequencing to characterize the virus' genome. When they compared it to other well-known filovirus genomes, they found that Lloviu virus represents a class of viruses distantly related to all ebolaviruses and that it may have diverged from ebolaviruses about 68,000 years ago.

"The detection of this novel filovirus in Spain is intriguing because it is completely outside of its previously described range. We need to ascertain whether other filoviruses native to Europe exist, and more importantly, if and how it causes disease," said Gustavo Palacios, the other first author of the study.

New ebolavirus-like filovirus found in Spain

Spanish and US scientists have discovered a new ebolavirus-like
filovirus in Spain, the 1st known detection of such a virus outside
sub-Saharan Africa and the Philippines. Reporting in PLoS Pathogens
yesterday [20 Oct 2011], they describe identifying the virus in a
specimen from a dead common bent-wing bat (_Miniopterus schreibersii_)
during an investigation of bat die-offs in southern Europe. They named
the novel pathogen Lloviu virus (LLOV) after the Cueva del Lloviu
region in which the bat was found. They write, "LLOV is genetically
distinct from other marburgviruses and ebolaviruses and is the 1st
filovirus detected in Europe that was not imported from an endemic
area in Africa." Filoviruses are among the most lethal of pathogens in
primates, including humans.

MARV was discovered in 1967 in Marburg, Germany during an outbreak in

With the exception of REBOV, that appears to be pathogenic in nonhuman

Bats are implicated as reservoirs and vectors for transmission of
filoviruses in Africa. ZEBOV sequences have been found in fruit bats
(_Hypsignathus monstrosus_, _Epomops franqueti_, and _Myonycteris
torquata_). MARV sequences have been found in fruit (_Rousettus
aegyptiacus_) and insectivorous (_Rhinolophus eloquens_ and
_Miniopterus inflatus_) bats. Bats naturally or experimentally
infected with ZEBOV or MARV are healthy and shed virus in their feces
for up to 3 weeks."

filovirus tropism for bats. However, unlike MARV and EBOV, where
asymptomatic circulation is posited to be consistent with evolution to
avirulence in this long-term host-parasite relationships, several
observations suggest that in the case of LLOV, filovirus infection may
be pathogenic. LLOV was found in the affected bat population (_M.
schreibersii_) but not in other healthy _M. schreibersii_ or in bats
of other species that cohabited the same caves (_M. myotis_)."

"Phylogenetic analysis of conserved domain III of the RNA-dependent
RNA polymerase confirms that LLOV belongs to the _Filoviridae_ and may
represent a complex of viruses related to all EBOVs. Phylogenetic
analysis of complete genome sequences (roughly 21 800 nucleotides)
confirmed that LLOV is a distinct genetic lineage that originates
after MARV."

"Recently, the discovery of integrated filovirus elements has led to
the proposal that filoviruses have co-evolved with mammals over
millions of years. Phylogenetic analyses of LLOV indicate a common
ancestor of all filoviruses at least 150 000 years ago."

"The sudden outbreak of bat die-offs in Spain that precipitated this
study destroyed several bat colonies in less than 10 days. Although we
have not demonstrated a causal relationship between LLOV and mortality
in _M. schreibersii_, the discovery of a novel filovirus in western
Europe, and the wide geographical distribution of the associated
insectivorous bat is a significant concern. While the virus was
detected in the north of Spain, simultaneous bat die-offs also have
