Manhattan, Kan. -- A veterinary professor and fellow researchers at Kansas State University?s College of Veterinary Medicine are conducting studies into the characteristics of the pandemic H1N1 flu virus and developing better detection methods.

Juergen Richt, a Regents Distinguished Professor in the veterinary college and a Kansas Bioscience Authority Eminent Scholar, is leading in vitro research to develop better H1N1 testing tools and overall preparation should the virus spread to swine populations, with a goal of protecting the swine industry. His group already has learned that H1N1 does infect pigs and will transmit between the animals but is not fatal.

The diagnostic tools they are working on would detect the virus directly by finding nucleic acids or other parts of the virus in samples, and indirectly by detecting antibodies produced by infected animals.

The researchers also studied the virulence of two strains of H1N1 in a nonhuman primate model to determine how the strains would affect humans. They found that an isolate strain from California was more virulent than one from Mexico, and that both are more virulent than seasonal H1N1 flu viruses.

Creating animal models for pandemic H1N1 is important, according to Richt, because physicians have two types of antiviral medications, one that targets the M2 protein and the other, including the drug Tamiflu, which targets the neuraminidase protein. Pandemic H1N1 has become resistant to the M2 inhibitors but is still sensitive to Tamiflu, Richt says. However, some pandemic flu isolates from humans are beginning to show resistance to Tamiflu.


The findings show the importance of research into zoonotic diseases, Richt says. ?Our strength at K-State is that we are very familiar with zoonotic diseases and that expertise is very critical now that an agent causing a pandemic flu in humans most likely originated in animal populations,? he adds. Zoonotic diseases will be a focus of the new $563 million National Agro and Bio-Defense Facility (NABF that is coming to Manhattan, Kan., replacing the government?s aging research facility at Plum Island, N.Y. The Manhattan site was chosen earlier this year over four other proposed sites.

Caution: it is a human virus. Pigs in Mexico are not responsible for the break. This H1N1 could be in human population for decades according to a recent PNAS study. It does infect pigs, but is mild (2009 JGV). Afterall, it is not virulent. will see what Richt and Webby can generate...BTW, the NBAF could be stopped by congress based on a recent news...

Fully agree, but these types of studies/opinions will get a lot of play from the swine lobby. The only way to put it to rest is to find, through seroprevalence studies probably, the origination (much of the evidence of which currently points to swine in the Far East).

Just to be 100% clear, Mixin is discussing "classic" H1N1 swine flu (.....deleted.....), which is different than "novel" human H1N1 (2009) aka "human swine flu".

There are 144 sequences of "classic" swine H1N1 in the NCBI database, starting in 1930.

A Riley County child has been infected with a strain of swine influenza not commonly seen in humans, but has fully recovered following a mild illness.

No other cases have been identified, but an investigation is underway.
The influenza strain that infected the child was identified as an H3N2 virus that commonly circulates in pigs in North America. It is different from the pandemic H1N1 virus, also of swine origin, that was first detected in the United States in mid-April.

"It is critical for people to understand that this H3N2 virus is not related to the pandemic H1N1 virus," said Jason Eberhart-Phillips, Kansas State Health Officer and Director of Health at the Kansas Department of Health and Environment (KDHE). "This is not a mutation or a recombination of the pandemic strain, and it does not appear at this time to be a threat to human health."

The child was likely exposed to the virus during the Riley County Fair in late July, where the child had direct contact with pigs. The child later developed influenza-like symptoms and sought medical care. The child has fully recovered and no other family members have reported illness.

KDHE is working closely with the Centers for Disease Control and Prevention (CDC), the U.S. Department of Agriculture (USDA), the Riley County Health Department and the Kansas Animal Health Department to investigate this case.

KDHE and the local health department are working to determine if the Riley County Fair swine exhibitors, or their pigs, have been ill.

Yes, and I cannot verify that information at all. I think they misspoke. Perhaps what was meant is that since 2005, 14 cases of human infection with swine flu have been detected. If it's 14 in 2009, well, then Houston, we have a problem.

I deleted a phrase here that made the my explanation confusing.

What I was trying to do was add detail to the terms to clarify whether "Classic" or "novel" swine flu was being discussed in each case. The original source could have been more explicit to decrease confusion.

Someone may have made a statement regarding a current cumulative total that a reported mistook for an annual total.

Since the only mention in the article about H1N1 was the pandemic strain, I assumed this quote was "mistakenly" referring to the pandemic strain.

This name thing is a real mess, imo. Nothing to do about it now; but can you imagine someone a hundred years from now combing through all the historical data trying to decide what flu they are talking about? We are in the here and now and have a hard time keeping it all straight. I guess for history's sake, we can be gratrful each pandemic had a distinct name.

After 2 years of tracking bird flu, My best friend is having problems making the mental switch and persists in calling panflu *bird* flu :D

I bet it's been happening alot more often because they are being alot more cautious. They stated "since 2005", and I find that interesting because that's when avian flu was quite popular, and now, I mean if this girl was hardly sick and recovered, I don't think they would have even tested her if there wasn't 2009 H1N1 around.

There were some guys from a pig farm near me who tested positive for a strain a month or so ago (Saskatchewan). They all recovered and didn't spread it. I bet that happens more often than we think. Of course it was recognized in them because of the constant testing we are doing now for influenza.

That's similar to two bird biologists who were tested for bird flu when they had mild flu, .... it turned out they had an H12 infection - what a surprise that was. Never would have been found if they hadn't been looking for H5N1. ;)

I think there is the cH1N1 and also the rH1N1 in swine (reassortant H1N1) and the rH1N1 might be what they are talking about below. (Please correct that if I'm wrong.)

The rH1N1 sounds harder to deal with in pigs than cH1N1 was.

Triple-Reassortant Swine Influenza A (H1) in Humans in the United States, 2005–2009


"Background Triple-reassortant swine influenza A (H1) viruses — containing genes from avian, human, and swine influenza viruses — emerged and became enzootic among pig herds in North America during the late 1990s.

Methods We report the clinical features of the first 11 sporadic cases of infection of humans with triple-reassortant swine influenza A (H1) viruses, occurring from December 2005 through February 2009, until just before the current epidemic of swine-origin influenza A (H1N1) among humans. These data were obtained from routine national influenza surveillance reports and from joint case investigations by public and animal health agencies.

Results The median age of the 11 patients was 10 years (range, 16 months to 48 years), and 4 had underlying health conditions. Nine of the patients had had exposure to pigs, five through direct contact and four through visits to a location where pigs were present but without contact. In another patient, human-to-human transmission was suspected. The range of the incubation period, from the last known exposure to the onset of symptoms, was 3 to 9 days. Among the 10 patients with known clinical symptoms, symptoms included fever (in 90%), cough (in 100%), headache (in 60%), and diarrhea (in 30%). Complete blood counts were available for four patients, revealing leukopenia in two, lymphopenia in one, and thrombocytopenia in another. Four patients were hospitalized, two of whom underwent invasive mechanical ventilation. Four patients received oseltamivir, and all 11 recovered from their illness.

Conclusions From December 2005 until just before the current human epidemic of swine-origin influenza viruses, there was sporadic infection with triple-reassortant swine influenza A (H1) viruses in persons with exposure to pigs in the United States. Although all the patients recovered, severe illness of the lower respiratory tract and unusual influenza signs such as diarrhea were observed in some patients, including those who had been previously healthy."

Say, does anyone know if the NBAF plan changes have anything to with this lab escape described below? Maybe the funding isn't there for a BSL 4 facility since they proved that you need that to work safely with swine flu viruses.

Two people developed symptoms of influenza 36 h after collecting nasal swabs from pigs experimentally infected with A/Sw/IN/1726/88 (Sw/IN). Pharyngeal swabs from these persons tested positive for influenza virus RNA 8 days after infection. Analysis of hemi-nested polymerase chain reaction (PCR) products indicated that the hemagglutinin (HA) segments of the isolates were genetically related to the HA of Sw/IN. Four influenza A virus isolates (A/WI/4754/94, A/WI/4756/94, A/WI/4758/94, A/WI/4760/94) were recovered from a 39-year-old man and 2 (A/WI/4755/94, A/WI/4757/94) from a 31-year-old woman. The HAs of the isolates were antigenically indistinguishable from the virus used to infect the pigs. Sequence analysis of the HA genes indicated they were 99.7% identical to the HA of the virus used in the experiment. Multisegment reverse transcription-PCR proved that all of the segments originated from Sw/IN, demonstrating that transmission of swine H1N1 viruses to humans occurs directly and readily, despite Animal Biosafety Level 3 containment practices used for these experiments.


Then not long after that escape...the novel (and troublesome) H3N2 popped up in that same geographical area, (that I think is now in the swine rH1N1 and the human pandemic H1N1 - please correct that if need be.)

In 1998, a novel H3N2 reassortant virus emerged in the United States swine population. We report the interspecies transmission of this virus to turkeys in two geographically distant farms in the United States in 2003. This event is of concern, considering the reassortment capacity of this virus and the susceptibility of turkey to infection by avian influenza viruses. Two H3N2 isolates, A/turkey/NC/16108/03 and A/turkey/MN/764/03, had 98.0% to 99.9% nucleotide sequence identity to each other in all eight gene segments. All protein components of the turkey isolates had 97% to 98% sequence identity to swine H3N2 viruses, thus demonstrating interspecies transmission from pigs to turkeys. The turkey isolates were better adapted to avian hosts than were their closest swine counterparts, which suggests that the viruses had already begun to evolve in the new host. The isolation of swine-like H3N2 influenza viruses from turkeys raises new concerns for the generation of novel viruses that could affect humans.
