A similar strain has already been detected in dead swans found last month and earlier this month at the Towada lakeside in neighboring Akita Prefecture and the shores of Lake Saroma in eastern Hokkaido.
Genetic tests have showed that the H5N1 strain, detected in Akita Prefecture and Hokkaido, are very close to those detected in poultry in South Korea.
In Tokyo, the Environment Ministry said it will make a genetic analysis of the H5N1 strain detected in the swans in Aomori Prefecture in an effort to determine links, if any, among the viruses detected so far.
Among the two swans in Aomori Prefecture was one which was found weakened on April 18 at the premises of a hotel on Towada's southern lake side. It died two days later. The other was one found dead on May 8 in the northern part of Towada.
Both swans tested negative for avian influenza virus in a preliminary examination. But the deadly H5N1 strain was detected later in detailed checks by Tottori University conducted on request by the Environment Ministry, prefectural government officials said.
Separate from the two in Aomori Prefecture, another dead swan was found last Sunday in the village of Sai on the northern tip of the Shimokita Peninsula. It will be sent to Tottori University next week for detailed virus checks, the officials said.
Meanwhile, the Environment Ministry said it has conducted virus checks on some 100 wild birds captured at Lake Towada between May 5 and 8, and some 100 sets of droppings from wild birds at Lake Saroma on May 12. They tested negative for avian influenza virus.
Medical experts say H5N1, an avian disease, may mutate or reassort into a strain capable of efficient human-to-human transmission and that an outbreak of avian influenza could kill millions.
The Japanese government has continued efforts to build up stockpiles of a Swiss-developed anti-flu drug called Tamiful.

The above translation describes the detection of H5N1 in two more whooper swans this week. They were in the same Towada Lake where the first positives were found a month ago (see satellite map). These swans were on the east side of the lake and therefore in the Amori Prefecture. A new outbreak in Korea was also reported this week, suggesting a new wave of birds may be migrating through the area.

The sequences of two isolates collected last month were virtually identical. There were Fujian (clade 2.3) reassortants with an HA related to clade 2.3.2. The other seven gene segments were related to clade 2.3.4. The NA sequence is most closely related to a human isolate from Guangdong province in 2006. The HA is closely related to a wild bird sequence from Hong Kong in 2007 which has been selected as the clade 2.3.2 vaccine target, but the sequence has been withheld. It is not clear of the withholding is linked to publication, or if the publication centers on the fact that the isolate is a reassortant. However, both isolates from Japan are reassortants, and the identity level of 99.7% or higher suggests the isolates in Korea are also Fujian reassortants linked to wild birds in Hong Kong..

Fujian sequences have not been reported previously in long range migratory birds like whooper swans. The detection in Japan over a one month period suggest that the level of Fujian infections is high, which is supported by the record levels reported in Korea, and the first reported H5N1 case in Primore in southeastern Russia.

The repeated detection of H5N1 in whooper swans in multiple locations over a one month time period raises additional concerns regarding an expanded geographical reach of H5N1. The current expansion in northeastern Asia involves the Fujian strain which has been more commonly reported in southern China and southeast Asia. Prior to the lab confirmation of H5 in the soldier in South Korea, the human Fujian isolates had been limited to the large clade 2.3.4 sub-clade. Since the 2007 / 2008 wild bird sequences in Hong Kong have not been released, it is not clear when the 2.3.2 / 2.3.4 reassortant formed, but its presence in long range migratory birds in locations in Japan and Russia in regions which have not previously reported H5N1 raise concerns of a significant global expansion of Fujian H5N1, which includes migration to North America.

The above translation describes additional dead or dying whooper swans in the Akita prefecture. Although initial rapid tests are negative, both of the recent confirmed positives in Aomori initially tested negative with the rapid test, which lacks sensitivity.

If these birds test positive they will extend the geographic reach of Fujian H5N1 in northern Japan (see satellite map) and increase the likelihood that the long range migratory birds represent a global expansion of the Fujian strain (clade 2.3.2 / 2.3.4) of H5N1.

Last month H5N1 was confirmed at three distinct locations in northern Japan, and the most recent confirmed positives have extended that reach.

Japan has acknowledged the shortcomings of the rapid test and has sent samples for more sensitive testing. The H5N1 in whooper swans in Japan are closely related to the H5N1 in Korea, which has led to the culling of over 8 million birds.
