Can Immunohistochemistry to Diagnostic Test AI

Avian influenza (AI) is one of infectious animal diseases that cause tremendous economic impact. Many believe, the cause of human bird flu infection is poultry, although the transmission is not known for certain. This was conveyed, a staff researcher at the Center for Veterinary Yogyakarta, DVM MSc Tjahyowati Swift.

Diagnostic methods have not found a fast and accurate for the detection of the virus in the field for prevention of transmission to humans can be done immediately.

Until now, there are 5 laboratory test that can be used to diagnose infection by AI virus, of whom an agent identification test with the method of isolation and identification of the virus in chicken eggs germinate. Then test the pathogenicity test intra-venous pathogenicity index (IVPI) at 4 weeks old chickens, serological test to detect the presence of antibodies in the blood of poultry, antigen detection test using a commercial rapid test kits and molecular tests to detect RNA.

Recently, Swift Tjahyowati successfully developed immunohistochemical test as a routine diagnostic test of AI. ''This method is very fast, accurate and secure, this test could be applied in the eradication of AI disease,''he said in an open examination to obtain his doctorate at the Faculty of Veterinary Medicine, Gadjah Mada University on Thursday (27 / 1).

Alumnus vet GMU Faculty of Veterinary Medicine in 1983, said immunohistochemical method (CPI) is a special staining technique to detect konstituent immunological tissue (antigen)''in situ''by using antigen-antobodi. ''It's just for our antibody material is still imported, and the price is relatively expensive,''he said.

Based on research results, test the streptavidin-biotin CPI can be used as a method of selection test for the detection of AI virus in poultry samples that have been fixed in formalin.

He said that the test technique can be used to indicate the existence of viral antigens on infected tissue as an indicator of viral involvement in the disease process so that it can be used to detect the presence of antigens of a microorganism that infects cells. Although the changes caused by microorganisms has not been seen either in macroscopic or microscopic.

The organs are recommended as an organ of choice for detection of AI virus samples to test the CPI, he said, are the organs that display pathological lesions of the brain and subsequent anatomic especially kidneys, lungs, spleen, liver, pancreas and heart.

''This test can be applied as a routine diagnostic method of AI virus infection in the Central Veterinary Laboratory Veterinary Wates and other equivalents,''he said.
