BEIJING, April 4 (Xinhua) -- China's agricultural authorities said Thursday the infectious H7N9 avian flu virus has been detected from pigeon samples collected at a marketplace in Shanghai, as the country reported its fourth death from H7N9 infection.

The samples were collected at a marketplace selling agricultural products in the Songjiang District of Shanghai and tested H7N9 positive by the national avian flu reference laboratory, according to a statement by the Ministry of Agriculture.

After gene sequence analysis, the national avian flu reference laboratory concluded that the strain of the H7N9 virus found on pigeons was highly congenetic with those found on persons infected with H7N9 virus.

The ministry has ordered beefed-up monitoring of H7N9 bird flu virus in more areas.

Also on Thursday, three more suspected H7N9 cases were reported in Shanghai, where three H7N9 human infection cases have been confirmed.

China has thus far confirmed 11 human cases of H7N9 infections.

China's health authorities have promised transparency and cooperation to the World Health Organization (WHO) in regards to human infections of the new strain of bird flu.

The Chinese Center for Disease Control and Prevention said Wednesday that no human-to-human transmission of H7N9 has been discovered and no epidemiological connection between these cases has been found.

A report appearing on China’s Ministry of Agriculture(MOA) website this morning has Twitter, and much of Flublogia abuzz this morning, as it indicates that at least one pigeon in Shanghai has tested positive for the emerging H7N9 virus.
While certainly an intriguing (and potentially important) development, this is simply another clue in this outbreak.

While this may indeed be a `smoking pigeon’, in truth we don’t know the mode of transmission to – or from – this bird – or how many other birds may be infected.

These are still early days, and one shouldn’t invest too heavily onto any one theory or scenario - no matter how plausible it may sound.

That said, a connection to live-bird markets might help explain the sudden, and fairly widespread emergence of the H7N9 virus in Eastern China.

.....Authorities still don't know what animal reservoirs harbor the virus, but one clue emerged today in the Chinese media, which reported that the virus had been detected in samples from pigeons that were at a Shanghai market.

Xinhua, citing the country's agriculture ministry, said the samples were collected at an agricultural marketplace in Shanghai's Songjiang district. Tests at China's national avian influenza reference laboratory found that gene sequences in the H7N9 viruses from the pigeons matched those that infected humans.

It said the ministry ordered more intensive monitoring for the virus. A Xinhua story posted later in the day said the pigeon findings prompted Shanghai authorities to close a live-poultry-trading zone and begin slaughtering all birds.

Officials also closed poultry trading areas in two markets in Shanghai's Minhang district after H7 viruses were detected in samples, according to the Xinhua report. The report did not specify what animals or items were sampled.

Regarding responses of pigeons to inoculation with NP CK/PA H5N2 or NP emu/TX H7N1, the pigeons remained clinically healthy throughout the 21-day observation period and did not have detectable levels of antibodies on 21 DPI; only one pigeon yielded the NP emu/TX H7N1 on 3 DPI. The virus was isolated from a tracheal swab and was believed to be the residual inoculum virus. Based on the responses of pigeons to NPAIV and HPAIV, it was concluded that the pigeons were resistant or minimally susceptible to infection with HPAIV or NPAIV.

Abstract

This investigation assessed the susceptibility of experimentally infected pigeons to the highly pathogenic avian influenza virus (HPAIV) H5N1 that caused recent outbreaks of avian influenza in birds and humans in several countries of Asia. For this purpose 14 pigeons were infected ocularly and nasally with 108 EID50 and clinical signs were recorded and compared with five chickens infected simultaneously as positive controls. The chickens demonstrated anorexia, depression, and 100% mortality within 2 days postinoculation. Three of the pigeons died after a history of depression and severe neurological signs consisting of paresis to paralysis, mild enteric hemorrhage, resulting in a mortality of 21%. Gross lesions in these pigeons were mild and inconsistent. Occasionally subcutaneous hyperemia and hemorrhage and cerebral malacia were observed. Microscopic lesions and detection of viral antigen were confined to the central nervous system of these pigeons. In the cerebrum and to a minor extent in the brain stem a lymphohistiocytic meningoencephalitis with disseminated neuronal and glial cell necrosis, perivascular cuffing, glial nodules, and in one bird focally extensive liquefactive necrosis could be observed. The remaining nine pigeons showed neither clinical signs nor gross or histological lesions associated with avian influenza, although seroconversion against H5 indicated that they had been infected.

These results confirm that pigeons are susceptible to HPAIV A/chicken/Indonesia/2003 (H5N1) and that the disease is associated with the neurotropism of this virus. Although sentinel chickens and most pigeons did not develop disease, further experiments have to elucidate whether or not Columbiformes are involved in transmission and spread of highly pathogenic avian influenza.

This is the first time that China has admitted to H7N9 in chickens.

It is curious that this result is made available only after worldwide speculation about the human spread of H7N9.

Chicken samples in Shanghai were revealed last night to be infected with the H7N9 virus.

It prompted the agriculture ministry to increase checks for the deadly strain of bird flu as the city temporarily closed live poultry markets and suspended live bird sales elsewhere.

The news came after the disease claimed its sixth mainland victim, while tests on the first suspected case in Hong Kong proved negative.

The ministry said last night that 10 chicken samples from two markets in the Minhang district of Shanghai and at the Huhuai Farm Products Market in the adjacent Songjiang district had tested positive for H7N9.

The virus was also found in two pigeon samples and seven environmental samples collected from these markets, out of a total 738 samples tested.

Hopefully we'll see some avian sequences soon to examine the transitions between the bird market animals and the human cases.
