Vaccine kick-starts immune response to cancer

CHICAGO (AFP) — Researchers have developed a plant-based cancer vaccine capable of kick-starting the body's immune response and being tailored to a patient's specific tumor type, according to a study.

While they have not yet determined whether the immune response is sufficient to destroy the cancer, the researchers are hopeful that the technique could one day lead to a cure for at lease some types of the deadly disease.

"This would be a way to treat cancer without side effects," said senior author Ronald Levy of the Stanford University Medical Center.

"The idea is to marshal the body's own immune system to fight cancer," Levy said Monday, adding that he's optimistic he'll get positive results from the next clinical trial.

"We know that if you get the immune system revved up, it can attack and kill cancer."

The process has already successfully cured cancer in mice.

Levy's team tested the vaccine on 16 patients who were recently diagnosed with follicular B-cell lymphoma, a chronic, incurable disease.

None of the patients experienced any significant side-effects and more than 70 percent of the patients developed an immune response, the study published in the Proceedings of the National Academy of Sciences found.

This is the first time a plant-based cancer vaccine has been tested on humans. There have been a few trials of cancer vaccines developed with animal or human cells but they have had mixed results.
The plant-based vaccine has a number of advantages.

It can be developed much more quickly and at far less expense. It does not carry the risk of infection should the animal cells be contaminated.

And the antibodies produced may also spark a stronger immune response than those developed in mammalian cells.

"Every lymphoma patient has a target on their tumor cells but each patient's tumor has a different version of that target," Levy said in a telephone interview.

Finding the right target requires cloning the genes from the patient's tumor.

Those genes are then injected into a virus which naturally attacks tobacco plants. This virus is scratched onto the leaves of a tobacco plant and it becomes a "protein production factor," Levy said.

A week later, the leaves are ground up and then the protein is isolated and injected into the patient.

"This technology is special because it's fast and very suitable to this customized, personalized approach because each plant can be making a different person's (vaccine)," Levy told AFP.

The vaccine would not be suitable for preventative purposes, Levy cautioned.

But the same technique could one day be used to fight other diseases, Levy added.

"We use proteins for a lot of purposes in medicine like enzyme replacement and vaccines and antibodies," he said.

"Growing human cells for production of proteins is really expensive and a long process and it's hard to maintain. Growing these plants is dirt cheap... and we know how to grow tobacco plants really well."

July 21, 2008 -- Plants may be a powerhouse for researchers making personalized vaccines for patients with follicular lymphoma, a type of non-Hodgkin's lymphoma.

The basic idea is to use plants as a factory to quickly and inexpensively grow vaccines tailored to each patient's follicular lymphoma.

That approach worked and was safe in a small, preliminary test noted in Proceedings of the National Academy of Science.

They tested their strategy on 16 follicular lymphoma patients, growing their personalized vaccines in tobacco leaves for three to four months.

Starting about six months after their last round of chemotherapy, the patients got their vaccine in a monthly shot, delivered to their thigh, every month for six months. Some also got shots of a chemical that boosted their immune response.

The point of the study was to see if the plant plan was practical and safe. It was; no side effects were reported and the plants grew the vaccines without messing them up.

More than 70% of the patients had an immune response to their vaccine and 47% had the specific immune response that was sought. But the study wasn't designed to test the effectiveness of the plant plan; further research is needed to see how well those vaccines work.

The researchers included A. A. McCormick of Large Scale Biology Corporation in Vacaville, Calif., which made the vaccines.

Tobacco 'could help treat cancer'


The tobacco plant - responsible for millions of cancer cases - may actually offer the means to treat one form of the disease, a study suggests.

US scientists used the plant to "grow" key components of a cancer vaccine.

The National Academy of Sciences study suggests they could be used to tackle a form of lymphoma.

UK specialists said while "potentially exciting", more research would be needed to test how well the vaccine actually worked.
The ironic new role for tobacco is the work of researchers from Stanford University in California.


They are using the plants as factories for an antibody chemical specific to the cells which cause follicular B-cell lymphoma, a type of non-Hodgkin's lymphoma.

These antibodies are put into a patient newly-diagnosed with the disease, to "prime" the body's immune system to attack any cell carrying them.

If successful, this would mean the body would then recognise and destroy the lymphoma cells.

However, every patient's antibodies are different, and would need to be produced quickly once the diagnosis was made.

The idea is not a new one - attempts have already been made to grow these antibodies inside animal cells, with mixed success.

However, a plant-grown vaccine would be much cheaper and in theory could carry less risk to the patient, as animal cells might hold unknown viruses.

So far, the experimental vaccine has only been tested on a handful of patients to check for any side-effects of using plant-produced antibodies, so its effectiveness at fighting the disease is uncertain.

'Infected' plants

Dr Ronald Levy, who is leading the research, said: "It's pretty cool technology - and it's really ironic that you would make a treatment for cancer out of tobacco. That appealed to me."

The technique is relatively straightforward. Once a patient's cancer cells are isolated in the laboratory, the gene responsible for producing the antibody is extracted and added to the "tobacco mosaic virus".

The plants are then "infected" with the virus, and as it spreads through the cells, the added gene starts the process of producing large quantities of the antibody.

After just a few days a few leaves are taken, ground up, and the antibody extracted from them.

Only a few plants are needed to make enough vaccine for a patient.

Professor Charles Arntzen, from Arizona State University, said that the sheer speed of the production process could convince patients to wait for their own tailored vaccine rather than undergoing other treatment.

A spokesman for Cancer Research UK, said: "While these results could potentially be very exciting, this was a small and early-stage trial and it did not look at whether this vaccination strategy reduced the size of the tumours."
"This is a good foundation for future work, but a larger study will be needed to test the success of this plant-made antibody in fighting non-Hodgkin's lymphoma."
